Modem Mode connections

My superhub 3 is connected to a TP-Link M4 router and is in Modem Mode...

Can I connect my Laptop into one of the other 4 Ethernet ports on the Hub3 to give me a non-wifi connection? My desktop PC is already connected to the Ethernet port on the M4 router.

No - when in modem mode only one ethernet port is active and you are using that for your own router. All ethernet connections must come from that - either directly or via adding an unmanaged switch with more GB ethernet ports.  Good 5-8 port units are <£20 from the usual online places.  Use good Cat5e or better ethernet cables.
